人血清白蛋白共振Rayleigh光散射检测氟离子

【摘要】 通过研究F- 离子对人血清白蛋白(HSA)溶液共振Rayleigh光散射(RLS)的增强效应 ,建立一种基于生物蛋白RLS增强测定环境中氟离子的新型检测方法。实验条件为λex=λem=368nm ,pH=5.80 ,t=32℃。结果表明 ,应用含氟离子溶液与空白溶液在368nm处RLS差值(ΔI) ,标准曲线法测定样品中的氟离子 ,方法线性范围为4.78~16.2μg/mL ,r=0.992 ,检出限为1.43μg/mL。方法精密度(RSD为1.31%~5.72 %)和准确度(回收率R为91 %~109 %)较好

【Abstract】 A novel method for the determination of fluoride ion(F - )with human serum albumin(HSA)was developed.It was based on the enhancement effect of F - on the resonance Rayleigh light scattering(RLS)of HSA in K 2 HPO 4 -NaH 2 PO 4 buffer solution.It was found that the enhanced intensity of RLS(ΔI)was proportional to the concentration of F - under the selected conditions:λ ex =λ em =368nm,pH=5.80,temperature=32℃,lay down time=30min.The linear range of the method was4.78~ 16.2μg/mLwith a correlation coefficient of0.992.The detection limit was1.43μg/mL.The RSD and the recovery were in the range of1.31%~5.72%and91%~109%,respectively.
