氨磷汀对化疗中外周血造血干/祖细胞的保护作用
The protective effects of amifostine on hematopoietic stem/progenitor cells against the chemotheraputic damage
【摘要】 目的 评估氨磷汀 (amifostine ,AMF)在保护造血干 /祖细胞免受化疗药物依托泊甙 (etoposide ,VP 16)损伤方面的作用。方法 提取新鲜和冻存的外周血造血干细胞 (PBSC)和HL 60细胞 ,分为AMF +VP 16组、VP 16组和空白对照组 ,用台盼蓝拒染法检测细胞活性 ,CFU GM集落培养计数 ,流式细胞仪测定细胞的凋亡率。结果 新鲜和冻存的PB SC样本中 ,AMF +VP 16组的细胞存活率和集落形成能力较VP 16组显著提高 (均P <0 .0 5 ) ;HL 60细胞样本中 ,AMF +VP 16组与VP 16组凋亡率的差异无统计学意义 (P >0 .0 5 )。结论 AMF能保护造血干 /祖细胞免受化疗药物VP 16的损伤 ,并且不影响VP 16对HL 60细胞的杀伤效果。
【Abstract】 Objective To study the protective effects of amifostine(AMF) on hematopoietic stem/progenitor cells against the damage of chemotheraputic drug—etoposide (VP、16). Methods Fresh and frozen peripheral blood stem cells (PBSC) and HL、60 cells were collected and divided into the AMF+VP、16 group,the VP、16 group and the control group. The cell viability,the number of CFU、GM and apoptosis were assessed. Results In fresh and frozen PBSC,cell viability and the number of CFU、GM of AMF+VP、16 group were both significantly higher than those of VP、16 group (all P< 0.05); In HL、60 cells,apoptosis of AMF+VP、16 group was a little higher than that of VP、16 group,but there was no statistical significance (P> 0.05). Conclusions AMF can significantly protect hematopoietic stem/progenitor cells against the damage of VP、16. Moreover,it does not reduce cytotoxity of VP、16 to HL、60 cells.
