日本血吸虫乳酸脱氢酶基因的获得和序列分析
Acquiring and Sequence Analysis of the Lactate Dehydrogenase Gene of Schistosoma Japonicum
【摘要】 目的 从日本血吸虫 (Schistosomajaponicum ,Sj)成虫cDNA文库中筛选日本血吸虫新基因 ,为日本血吸虫病的诊断或防治提供靶抗原。 方法 筛选日本血吸虫成虫cDNA文库 ,随机挑取重组阳性克隆进行DNA测序 ,再对新基因序列进行步移法测序获取全长cDNA ,并利用生物信息学技术对其进行分析。 结果 获得了 1个新基因 ,其cDNA全长 12 5 1bp(AY2 2 5 190 ) ,编码 3 3 1个氨基酸 ,编码蛋白与人类或螈、蟾蜍、鸡、鼠及猪等其它生物的乳酸脱氢酶高度同源。 结论 利用表达序列标签法、步移法测序和生物信息学技术三者相结合 ,成功获得了编码日本血吸虫乳酸脱氢酶基因的cDNA全长序列。
【Abstract】 Objective To acquire a new gene from an cDNA library of adult Schistosoma japonicum(Sj) and to search new target antigens for diagnosing and preventing and treating schistosomiasis. Methods A cDNA clone was isolated randomly from the cDNA library of adult Sj (Chinese strain) by expression sequence taqs(ESTs) stratagem, and then it was sequenced by primer-walking sequencing. A novel gene was acquired after analysis by BLAST programs and other programs in GeneBank databases. Results The gene(AY225190) code for lactate dehydrogenase of Schistosoma japonicum (SjLDH)was acquired, which had 1 251 bp and encoded 331 amino acids. The deduced protein sequence was highly homologous with LDH sequence of human, ambystoma, xenopus, chicken, rattus and pigeon. Conclusion The gene of SjLDH was successfully acquired by ESTs stratagem and primer-walking sequncing and bioinformatics technique.
