茶多酚对异丙肾上腺素所致小鼠心肌肥厚的影响
Effects of Tea Polyphenol on Cardiac Hypertrophy Induced by Isoprenaline in Mice
【摘要】 目的 观察茶多酚(TP)对异丙肾上腺素(Iso)所致小鼠心肌肥厚的抑制作用。方法 Iso 1 mg/kg,每日两次,连续皮下注射(sc)14 d,形成小鼠心肌肥厚模型,观察小鼠心电图变化,分别测定心脏各重量参数及血清超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、一氧化氮(NO)含量、肌酸磷酸激酶(CPK)含量和乳酸脱氢酶(LDH)含量。结果 Iso连续sc 14 d后,心电图提示左心室"肥厚劳损"。心重指数(HWW/BW、LHWW/BW)明显增加,血清SOD活性下降、MDA含量增加、NO含量下降、CPK和LDH含量增加。给TP 150、300、600 mg/kg和卡托普利100 mg/kg均可改善左心室肥厚后形成的"劳损"征象,降低心重指数,增加SOD活性,降低MDA含量,增加NO含量,同时降低CPK和LDH含量。结论 TP能抑制Iso引起的心肌肥厚,该作用与清除氧自由基及升高NO含量有关。
【Abstract】 Objective To study the effects of tea polyphenol (TP) on prevention of cardiac hypertrophy induced by isoprenaline (Iso) in mice. Methods Mice cardiac hypertrophy model was established by continued administration of Iso 1 mg/kg sc, twice per day for 14 days, the heart - weight -index, change in electrocardiogram and the contents of SOD, MDA, NO, LDH, CPK in serum were measured. Results After a continued administration of Iso sc for 14 days, ’cardiac hypertrophy fatigue’ was able to be seen from the electrocardiograms and the heart-weight-index was increased. Also, there was a decrease in SOD activity, an increase in MDA, LDH, CPK content and an increase in NO content were observed. Conclusion TP can inhibit cardiac hypertrophy induced by Iso and this effect may be related to eliminating the oxygen free radicals and raising NO level.
