乳腺癌C-erbB-2、P16基因蛋白表达与多药耐药的相关分析

【摘要】 目的 探讨乳腺癌中C -erbB - 2、P16与GST -Pi、TOPOⅡ、PgP耐药基因表达的相关性。方法 采用即用型微波加热免疫组化法 ,对 5 7例乳腺癌C -erbB - 2、P16、GST -Pi、TOPOⅡ、PgP进行检测分析。结果5 7例乳腺癌中C -erbB - 2、P16、GST -Pi、TOPOⅡ、PgP阳性表达率分别为 6 1.4 0 %、36 .84 %、5 2 .6 3%、2 4 .5 6 %、15 .79%。浸润性导管癌对 3种耐药基因有不同程度的阳性表达 ,较其它组织学类型的乳腺癌的耐药基因更为广基。乳腺癌C -erbB - 2、P16、GST -Pi、TOPOⅡ、PgP阳性表达与年龄、临床分期、腋下淋巴结转移无明显关系。结论 乳腺癌产生多药耐药性可能与C -erbB - 2、P16异常表达相关。C -erbB - 2、P16异常表达可能是乳腺癌产生多药耐药的因素之一。部分乳腺癌患者体内存在多项耐药基因或癌细胞内原发存在对GST -Pi、TOPOⅡ、PgP介导的抗癌药物已产生耐药性。

【Abstract】 Objective To explore the relationship between C-erbB-2 and P16 genes expression and expression of multidrug resistance genes, GST-Pi?TOPOⅡ and PgP.Methods Detection and analysis of C-erbB-2?P16?GST-Pi?TOPOⅡ?PgP gene proteins in 57 breast cancer patients using instant microwave heating-immunohistochemistry technique were performed.Results In the 57 cases of breast cancar patients, the positive expression of C-erbB-2?P16?GST-Pi?TOPOⅡ and PgP genes proteins were 61.40%,36.84%,52.63%,24.56% and 15.79% respectively.Abnormal expression of C-erB-2?P16 were associated with the positive expression of GST-Pi?TOPOⅡ and PgP genes. All invasive ductal carcinoma had different levels of protein expression of multidrug resistant related genes,GST-Pi?TOPO-Ⅱ and PgP.Moreover expression level and positive exprssion rates were higher than other tissue type of breast cancer.No relationship were found between expression of C-erbB-2?P16?GST-Pi?TOPO-Ⅱ and PgP genes and patient’s age, clinical stage, and axillas lymph nodes metastasis in this study. Conclusion Abnormal expression of C-erbB-2 and P16 genes are related to multidrug resistance in breast cancer.Further, overexpression of C-erbB-2 and P16 genes are probably the factor of multidrug resistance. Partial breast cancer patients have primary positive expression of GST-Pi?TOPO-Ⅱ?PgP genes,which are causing factors of intrinsic multidrug resistance.
