缬沙坦对阿霉素肾病肾硬化大鼠血小板源性生长因子表达的影响
Effects of valsartan on expression of platelet-derived growth factor in the kidney of rats with adriamycin-induced nephrtic glomerulosclerosis
【摘要】 目的 :探讨血管紧张素 - 1型受体拮抗剂 ( AT1RA)减轻肾小球硬化的机理。方法 :将 36只大鼠分为对照组、模型组和治疗组 ,模型组和治疗组行单侧肾切除大鼠加阿霉素注射诱导阿霉素肾病肾硬化模型 ,治疗组每日给予缬沙坦 2 0 mg/kg灌胃 1次 ,共 1 2周。免疫组化法检测肾组织血小板源性生长因子 - BB( PDGF- BB)的表达。结果 :模型组较对照组肾小球硬化明显 ,表达明显增强 ;缬沙坦治疗组肾小球硬化较模型组明显减轻 ,PDGF- BB表达亦减弱。结论 :阿霉素肾病大鼠肾小球硬化时 ,PDGF- BB表达增强并参与肾小球硬化的进展 ;AT1RA可能通过降低 PDGF- BB在肾组织表达发挥其延缓肾小球硬化的作用
【Abstract】 Objective:To inquire into the mechanism of angiotensinⅡ type 1 receptor antagonist(AT 1RA) valsartan in lightening glomerulosclerosis.Methods:36 rats were divided into control group, model group and treatment group. Uninephrectomy and injection adriamycin induced the rats model with glomerulosclerosis. Valsartan(20mg/kg) was orally administered by daily rats in treatment group for 12 weeks. Immunohistochemistry was utilized to detect the expression of platelet-derived growth factor-BB(PDGF-BB) and the level of PDGF-BB was hemi-quantitated.Results:Compared with those of the control group, the kidneys of the model group had more severe glomerulosclerosis and higher level of expression of PDGF-BB. The degree of glomerulosclerosis and the level of expression of PDGF-BB were improved by valsartan treatment.Conclusion:The level of PDGF-BB is higher in the kidney of rats with adriamycin-induced nephrtic glomerulosclerosis. PDGF-BB takes part in glomerulosclerosis. AT 1RA valsartan may suppress the expression of PDGF-BB so as to postpone the process of glomerulosclerosis
