蛇毒蛋白C激活剂实验条件及稳定性的探讨
Research on the Experimental Conditions and Stability of Protein C Activator Isolated from Snake Venom
【摘要】 目的 探讨可能影响蛇毒蛋白 C激活剂作用的实验条件和该试剂的稳定性。方法 分别采用APTT法和 CBS0 2 .46底物法 ,检测蛇毒蛋白 C激活剂在不同实验条件下的抗凝活性及酰胺酶活性。结果 蛇毒蛋白 C激活剂的最适 p H值为 6.0~ 7.0 ,最适温度为 37~ 40℃ ,肝素、枸橼酸钠等抗凝剂及 Na+ 、K+ 、EDTA等不影响其活性 ,SDS则显著抑制其生物学活性。该试剂溶液剂型在室温下或 4℃中 4w内效价无变化 ,冻干剂型则在 2 y内效价无明显变化。结论 蛇毒蛋白 C激活剂是一种效价较高、稳定性较好、影响因素少的蛋白 C活化试剂
【Abstract】 Objective To investigate the experimental conditions and stability that may affect the activities of Protein C activator(PCA-SV).Methods PCA-SV’s possible anticoagulant and amidase activities were measured by method of APTT and that of specific chromogenic substrate respectively in various experimental conditions.Results The pH-optimum and temperature-optimum for the action of PCA-SV were 6.0~7.0 and 37~40 ℃ respectively.The anticoagulants(heparin and Sodium Citrate) had no effect on the activities.SDS significantly inhibited the biological action of PCA-SV.In room temperature or 4 ℃,the titre had no change within 4 weeks for solution reagent and 2 years for dry reagent.Conclusion PCA-SV is a new protein C-activating reagent that has high potency and stability.
