银染PCR-SSCP检测卵巢癌TGF-β及Smads信号改变的意义

【摘要】 目的 通过检测卵巢癌 TGF-β介导的信号转导通路的受体、Smads基因突变 ,研究该信号转导在卵巢癌发生中的作用 ,揭示卵巢癌的发病机制。方法 收集卵巢癌手术切除标本 ,并以病人正常组织作为对照 ,提取 DNA,依据待测基因的核心高突变区设计引物 ,采用银染 PCR- SSCP法检测 TβRI、TβR 、Smad2及 Smad4基因突变。结果  TβR ( 1 6/46)、TβR ( 2 9/46)、Smad2 ( 1 8/46)在卵巢癌中有较高突变率 ,Smad4( 1 /46)仅零星突变。结论  TGF-β介导的信号转导途径在卵巢癌的发生中起重要作用。

【Abstract】 Objective With the goal of interpreting the association of ovarian carcinomas with signal transducting mediated by TGF-β and Smad family,mutations in the components of the signaling were analyzed by PCR-SSCP,which would contribute to the understanding of the occurrence of ovarian carcinomas.Methods Resected samples of ovarian carcinomas were collected,and DNA were extracted both from ovarian carcinomas and from the normal parts of the samples (as control).Specific mutant DNA were then propagated by PCR under appropriate primers of TβRⅠ,TβRⅡ,Smad2 and Smad4.The productions of PCR were analyzed by single strand conformation polymorphism (SSCP).Results The analysis showed that mutations of TβRⅠ(16/46),TβRⅡ(29/46) and Smad2(18/46) frequently harbored in ovarian cancer,but Smad4(1/46)mutation was seldom found.Conclusion TGF-β and Smad family signaling is involved in the occurrence of ovarian carcinomas.
