网翅蝗科(Arcypteridae)部分种类全基因组提取及16S rRNA基因和COⅠ基因测序
Extracting Genomic DNA and sequencing of 16S rRNA and CO Ⅰ genes in Arcypteridae portion species
【摘要】 通过实验提取中国甘肃境内网翅蝗科蝗虫全基因组DNA序列,探索了直翅目蝗虫的总 DNA提取方法;通过特异性引物,对线粒体16S rRNA基因和COⅠ基因序列进行了扩增,并通过 多次预实验探索这两个基因在网翅蝗科昆虫中扩增的最佳条件,为将来的网翅蝗科系统发育分析 奠定了实验基础.扩增好的样品用于测序,测序结果可用于分析6种蝗虫的系统发育关系.
【Abstract】 Mitochondrial DNA are advantages over nuclear DNA inestimating phylogenetic telationships among insect taxa because of it rapid evolution and inheritance without gene recombinations . 16S rRNA and CO1 DNA sequence data from the mitochondrial genome are widely used in insect molecular systematics. The paper present the process of the DNA extract and PCR . Deeply discussed the potential uses of these two gene.
