苏云金杆菌和环磷酰胺对中华稻蝗DNA的损伤

【摘要】 用微量注射处理法初步研究苏云金杆菌和环磷酰胺对中华稻蝗在分子水平上的毒性作用.在不同的药剂作用时间解剖虫体,取其精巢,低渗后固定保存,用传统的酚/氯仿法提取精巢总DNA(部分技术环节略作改动)电泳检测比较结果,并进行PCR随机扩增.结果显示,两种药物对中华稻蝗总DNA有明显的降解作用,苏云金杆菌处理的样品均没有随机扩增产物,环磷酰胺部分样品有随机扩增条带,但不同样品扩增条带之间的可比性较差.

【Abstract】 The DNA damage of Bacillus thuringiensis Berliner(B.t) and cyclophosphamide(CP) to Oxya chinensis in molecular level were studied.The treated grasshoppers were dissected from the ventral side in different timepoint,the testes were collected,immersed in 0.075 mol/L KCl and then fixed in 3∶1 methanol-glacial acetic acid.The total DNA was extracted with phenol-chloroform by the modified traditional methods in technology and the results were compared by electrophoresis of the agarose gel. In addition,Randomly Amplified Polymorphic DNA was used to preliminarily analyze the DNA damage effect of these two agents on O. chinensis. The results showed that the total DNA,which was extracted from the samples treated by B.t and CP,was seriously decomposed to little fragments. RAPDs of all the samples treated by B.t had no bands and the partial samples treated by CP obtained some bands,however,there were no significant comparability among these bands.
