产纤溶酶枯草杆菌液体发酵工艺优化研究
Optimization of Fermentation Process on A Fibrinolytic Enzyme from Bacillus subtilis SBS
【摘要】 采用单因子实验和正交实验对高产纤溶酶的枯草杆菌最佳发酵工艺进行了优化 ,结果表明 ,该菌株分泌的胞外酶具有较强的体外溶栓作用 ,产纤溶酶最佳的发酵条件为 :3%可溶性淀粉 ,2 %豆浆全汁 (鲜豆 ) ,0 .0 2 %CaCl2 ,培养温度为 37℃ ,初始pH8.3,装液量为 2 5 0mL三角瓶 5 0mL ,发酵时间 4 4h左右 ,Ca2 + 、Mg2 + 和Mn2 + 对酶活力有促进作用 ,Cu2 + 对酶活性有强烈抑制作用。
【Abstract】 The fermentation process producing a potent fibrinolytic enzyme from Bicillus subtilis strain SBS was optimized in shaking flask through single factor experiment and orthogonal experiment.The results showed that the extracellular enzyme produced by the strain SBS had a stronger thrombolytic action in vitro.The optimal fermentation condition as follow:3% soluble starch,2% soybean milk(fresh weight),0.02% CaCl2,initial pH8.3 and 50 ml culture medium in 250 ml flask at 37℃ for 44 hours.In addition,metal ions such as Ca2+,Mg2+ and Mn2+ can increase the enzyme activity,however,Cu2+ inhibit it violently under the same condition.
