均匀设计法对产几丁质酶细菌C4发酵条件的优化

【摘要】 系统研究了碳源 ,氮源 ,起始pH值、培养基装量、培养温度和时间等因素对细菌C4产几丁质酶的影响。结果表明 ,碳、氮源分别以胶体几丁质、KNO3 和蛋白胨最好 ;在起始pH值 7.6~ 8.5 ,培养基装量为三角瓶体积的 12 % ,培养温度 2 8℃ ,振荡培养 (180r/min) 5d时最有利于几丁质酶的产生。在此基础上通过均匀设计法优化了发酵培养基配方。优化后的培养基配方为 :胶体几丁质 1.5 % ,蛋白胨 0 .5 5 % ,KNO3 0 .3% ,MgSO40 .0 9% ,Tween80 0 .0 0 5 %。在该条件下 ,几丁质酶活力达 2 .6 8U/mL ,比在原基础培养条件下的酶活力提高 90 .1%。

【Abstract】 The effects of a number of factors on the production of chitinase were studied,including carbon source,nitrogen source,pH,volume of culture media,ultural temperature and time.The results showed that the optimal carbon source and nitrogen source were colloid chitin,KNO3,and peptone,respectively.The optimization pH 7.6~8.5,cultural temperature:28℃,and C4 was cultivated for 5d on 180r/min shaker in flask with 12% volume medium.On the basis of this,the recipe of fermentation medium was optimized through uniform-design.A suitable fermentation condition was found for chitinase-producing strain C4.The optimal conditions were as follows:colloid chitin 2.5%,peptone 0.55%,KNO3 0.3%,MgSO4 0.09%,Tween 80 0.005%.Chitinase activity reaches 2.68U/mL under the conditions,bove 90.1% that of the basic fermentation conditions previously.
