重组β酪蛋白磷酸肽二聚体基因的构建、表达及其生物活性分析
Construction, Expression and Bioactivity Assay of Recombinant β-CPP Dimer Gene
【摘要】 生物活性短肽 β酪蛋白磷酸肽 (β CPP)能够增加细胞对二价离子的吸收、促进受精过程并对细胞增殖产生影响 .为深入研究其生物学功能 ,采用基因工程方法得到β CPP的二聚体基因并构建真核表达载体 ,转染CHO细胞建立稳定表达细胞系 .鉴定纯化所表达产物 ,考察表达产物对精子细胞内游离Ca2 + 水平、细胞增殖和细胞凋亡等方面的影响 .结果表明 ,重组 β CPP二聚体能够显著增加精子细胞对Ca2 + 的吸收、促进脾细胞的增殖并诱导某些肿瘤细胞的凋亡 ,有着广泛的生物学活性和潜在的药理学意义
【Abstract】 Casein phosphopeptide (β-CPP) influences numerous biological processes such as uptake of the bivalent ions and proliferation of the cells. To further investigate biological functions of β-CPP, eukaryotic expression vector of β-CPP dimer was constructed and then transfected into CHO cells. After selection, the cell lines expressing β-CPP dimer were obtained and the recombinant products were identified and purified. Activity assay of the recombinant protein indicated that the recombinant β-CPP dimer could improve Ca 2+ uptake of sperm,stimulate the proliferation of spleen cells and induce apoptosis of some malignant tumour cells.
