藻酸盐水凝胶与前软骨干细胞构建组织工程化骺板
In vivo engineering growth-plate-like tissue by composite of alginate hydrogel and precartilaginous stem cells
【摘要】 目的探讨藻酸盐水凝胶(alginate hydrogel,AH)与前软骨干细胞(precartilaginousstem cells,PSCs)复合物体内构建骺板样组织的可行性。方法利用多聚赖氨酸和藻酸盐制备AH。将PSCs在含有TGF-β3的藻酸盐微珠中培养,分别于7、14、21天将被诱导的PSCs包埋入AH制备成AH-PSCs复合物,未被诱导的PSCs作对照。分别将AH-PSCs自体植入大鼠背部肌肉中,定期取材测定移植物的重量、细胞数,并行组织学检查及Ⅱ型胶原免疫组化检测。结果PSCs被诱导7天组移植物大约70%能形成骺板样结构;PSCs被诱导14天组及21天组的移植物能形成软骨组织及骨组织,但无骺板样结构;PSCs未被诱导的移植物无软骨组织及骨组织形成。被诱导7天组的移植物在20周的平均重量及每个移植物所含的细胞数均大于被诱导14天组或21天组的移植物(P<0.01);被诱导7天组的移植物在20周时平均重量及每个移植物所含的细胞数均大于在4周或10周时(P<0.01)。结论TGF-β3诱导PSCs7天的AH-PSCs复合物在体内能构建骺板样组织。
【Abstract】 Objective The goal of this study was to investigate the possibility of engineering growth-plate-like tissue by composite of alginate hydrogel and precartilaginous stem cells. Methods Alginate hydrogels (A-H) were prepared by poly-L-lysine and alginate. PSCs were cultured in alginate bead in presence of TGF-β3, and then differentiated PSCs were entrapped in AH at presence of TGF-β3 for 7, 14 or 21 days, respectively. The composites of AH and PSCs were autologously transplanted in dorsal muscles of SD rats. The harvested implants were measured and weighed, and then examined by histology, immunohistochemi stry of type Ⅱ collegen and the number of cells (according to the amount of DNA). Results Histologic evaluation of implats of TGFp3-inducing-7-day indicated that 70% of implants revealed macroscopic organization of the growth-plate-like structures. The implants of TGFβ3-inducing-14-day or TGFβ3-inducing-21-day group revealed formation of cartilage and bone, but not growth-plate-like structures. The implants of undifferentiated PSCs didn’t reveal cartilage-like or bone-like tissues. The mass and total number of cells in TGFβ3-inducing-7-day group was significantly greater than that in TGFβ3.-inducing-14-day or TGFβ3-inducing-21-day group at 20 weeks (P<0.01). Within TGFβ3-inducing-7-day group, the mass and total number of cells of implants was significantly greater at 20 weeks than at 4 or 10 weeks (P<0.01). Conclusion Transplantation of composite of alginate hydrogel and precartilaginous stem cells that induced by TGF-β3 for 7 days revealed macroscopic organization of the growth-plate-like structures.
