节育剂微胶囊的制备及其抗生育作用

【摘要】 以从印楝油氯仿提取物中分离纯化出的 2 十八酸 4 十六酸 2 ,4 二戊酯为主体药物 ,采用复凝聚法 ,将其制成平均粒径为 10 0~ 180 μm的微胶囊 ,考察了微胶囊的形态特征、包封率、载体回收率 ,并以昆明小鼠为试验动物 ,采用组织学和流式细胞术研究了该胶囊的抗生育作用。结果表明 ,该制剂能实现对药物的有效负载 ,包封率达到 90 %以上。该体系经过口服后 ,不影响其抗生育作用 ,可保持较长时期的可逆性不育。组织学和流式细胞术检测结果表明 ,该制剂的抗生育作用机制主要是破坏生精过程和杀精子作用。

【Abstract】 The main component of this pill is 2-Octadecanoic acid -4 -Palmitic acid-2,4-Pentanediyl ester separated from chloroform extract of ne em oil.The microcapsules coated by the re-curdle method were fabricated with a n average particle size of 100~180 μm. The morphological characteristics, inco rporation efficiency, carrier reclamation efficiency of the microcapsule were in vestigated. Kunming mice were used in the experiment,and the anti-fertility eff ect of the microcapsule on the histology and apoptosis was studied by light and electron microscopy and the flow cytometry. The data obtained clearly indicated that the microcapsule could lead to the payload of medicine, the incorporation e fficiency being 90%. After the microcapsules were given to the male mice orally, its anti-fertility effect came into being and could keep the mice in a state of reversible infertility for a long time. The results of histological study and f low cytometry indicate that the mechanism of its anti-fertility effect involve s mainly the inhibition of sperm motility and the arrest of spermatogenic proce ss.
