色谱法在植酸酶水解产物分离和鉴定中的应用
The application of HPLC in purification and determination of isomers of inositol mono- to hexaphosphates from phytate by extra-cellular phytases
【摘要】 利用离子对色谱、离子色谱方法对A.niger496-1菌株产生的胞外植酸酶系水解植酸产生的产物—低磷酸肌醇进行了分离和鉴定。实验结果表明离子对色谱可有效分离、鉴定IP3~IP6;离子色谱方法1可以有效的分离和鉴定IP2~IP6及IP3~IP5的异构体,离子色谱方法2可以有效测定IP1~IP3及它们的异构体,离子色谱方法1和方法2结合使用可以完全分离植酸酶水解植酸产物中所有的磷酸肌醇及它们的异构体。研究还表明A.niger496-1菌株产生的植酸酶对植酸水解的第一步具有很强的选择性,仅形成一种五磷酸肌醇。
【Abstract】 Ion-pair HPLC and high performance ion chromatograph(HPLC) methods for separation and determination of isomers of inositol mono- to hexaphosphate (IP1~IP6) from phytate by extra-cellular phytases of A.niger 496-1 are investigated.Experiments prove that IP3~IP6 in sample could rapidly be separated and determined by ion-pair HPLC,IP2~IP6 in it is determined by HPIC,which also separated isomers of IP3~IP5,and IP1~IP3 and their isomers could be seperated by HPIC.The experiment also confirms that the initial dephosphorylation of phytate by extra-cellular phytases of A.niger 496-1 is given only 1,2,4,5,6-IP5.
