GABA介导杏仁外侧核对皮层AⅠ区神经元声反应的抑制:在体微电泳研究(英文)

【摘要】 在SD大鼠上应用多管微电极方法,观察微电泳GABA及其受体的拮抗剂或激动剂对杏仁外侧核(LA)抑制皮层AⅠ神经元声反应效应的影响。结果显示,电泳GABA能抑制皮层AⅠ区神经元的电活动,电泳GABA_A受体拮抗剂bicuculline(BIC)则能易化其反应;电刺激LA能抑制皮层AⅠ区听神经元声反应,电泳GABA产生类似于刺激LA的抑制效应;LA对皮层AⅠ区神经元的抑制效应能被BIC所翻转,而不能被甘氨酸受体拮抗剂strychnine所翻转,电泳GABA_B型受体激动剂baclofen对神经元声反应无影响。上述结果表明,GABA可能是介导LA抑制皮层AⅠ区神经元声反应的最终递质,并且是通过GABA_A受体起作用的。

【Abstract】 Experiments were performed on Sprague Dawley rats with multibarrel microelectrode technique. The effects of acousticresponse of A Ⅰ cortex neurons produced by electrical stimulation of lateral amygdaloid nucleus (LA) and the influence of GABA wereobserved. Experimental results showed that iontophoretic administration of GABA caused a pronounced inhibition of the electricalactivity of A-Ⅰ neurons. Blockade of GABA_A with bicuculline (BIC) facilitated the acoustic response. The acoustic response of A-Ⅰneurons was inhibited when the LA was stimulated. lontophoretic application of GABA resulted in a similar inhibitory effect as that ofLA stimulation. Blockade of GABA_A with bicuculline reversed the inhibitory effect of LA stimulation on the acoustic response of A-Ⅰneurons. In contrast, application of strychnine, a glycine receptor antagonist, could not reverse the inhibitory effect of LA. Baclofen, aGABA_B agonist, did not affect the acoustic response of the auditory neurons. These results indicate that GABA is the ultimate transmitterwhich mediates the LA stimulation-induced inhibition of the acoustic response of A-Ⅰ neurons in rats, possibly via the GABA_A receptor.
