那西肽产生菌S.actuosus选育及发酵条件的优化
Breeding of S.actuosus and Optimize Fermentation Conditions
【摘要】 目的 提高那西肽产量。方法 介绍那西肽产生菌活跃链霉菌S .actuosus(Z10 ) 的诱变育种 ,诱变的方法有紫外线、亚硝基胍及两者的联合诱变 ,并对高发酵单位菌种优化发酵条件和测定其生化曲线。结果 通过紫外线及亚硝基胍联合诱变获得高产菌株Z12 ,该菌种进行 4次传代试验显示稳定 ,经发酵条件优化后在培养基中添加 2 %黄豆粉 ,0 .0 1%硫酸锰可以提高那西肽产量。菌种Z12 相对于出发菌种Z10 提高那西肽产量达 5 8.9%。结论 通过紫外线及亚硝基胍联合诱变可以筛选到高活力菌种
【Abstract】 Purpose To improve the production of Nosiheptide. Methods This research paper reported the breeding of S.actuosus(Z 10).The way of mutation included UV、NTG and UV-NTG.the fermentation medium was optimized and bio-curve was assayed. Results The mutagenesis of UV-NTG could effectively improve the nosiheptide production.The highest production strain Z 12 was stable after 4 regeneration.The optimal fermentation medium was added more 2% soybean and 0.01% MnSO 4.Comparing the strain of Z 10,Z 12 had higher yield of nosiheptide about 58.9%. Conclusions The mutagenesis of UV-NTG could effectively improve the nosiheptide production.
