华佳8号葡萄的离体繁殖技术

【摘要】 本文以葡萄品种“华佳8号”为试验材料,以茎段为外植体,进行愈伤组织、不定芽、不定根诱导试验。结果表明:诱导愈伤组织的最佳培养基为MS+BA2.0~3.0mg·L-1(下同,略)+IAA0.02~0.05,诱导率达到100%,其中BA3.00配合IAA0.05愈伤组织增殖速度最快;诱导不定芽的最佳培养基为1/2MS+BA3.00+IAA0.05,诱发率达90%以上;诱导不定根的最佳培养基为MS+BA0.10~0.20+IBA1.00,生根率达到90%。

【Abstract】 The vine variety “Huajia No.8”is a good rootstock, especially fo r the variety“Tengren”, but it is very difficult to form roots for cuttage . To improve rooting frequency in vitro propagation techniques were studied in this p aper. Results showed the best medium for callus formation frequency was MS+BA2. 0~3.0 +IAA0.02~0.05, its induction rate being 100%.The best medium for adven titious bud differentiation was 1/2 MS+BA3.0+IAA0.05,its differentiation rate being above 90%.The best rooting formation was observed on MS+BA0.1~0.20 +IB A1.0 , the rate being above 90%.At last, seedlings by in vitro propagation of t he vine variety “Huajia No.8”were obtained.
