菊花外植体再生体系的研究
STUDY ON REGENERATION SYSTEM OF CHRYSANTHEMUM EXPLANTS
【摘要】 以不同小菊品种的叶片、茎段、花瓣及子房为外植体进行再生培养。实验结果表明 :不同品种、不同外植体对相同激素水平的反应是不同的。以叶片为外植体时 ,品种Rm 2 7 2的分化率高于Rm 6 4 ;Rm 2 7 2最适培养基为MS +BA 0 .5mg/L +NAA 0 .5mg/L和MS +BA 2 .0mg/L +NAA 0 .2mg/L ,而品种Rm 6 4为MS +BA 6 .0mg/L +NAA 0 .4mg/L ;茎段分化率高于叶片 ,叶片基部分化率高于中上部。子房培养较适宜的培养基为MS +BA 1.0mg/L +NAA 1.0mg/L和MS +BA 2 .0mg/L +NAA 2 .0mg/L ;MS +BA 2 .0mg/L +NAA 0 .2mg/L培养基可有效诱导花瓣产生愈伤组织及芽的分化。并对细胞分裂素、生长素在再生中的作用及适宜浓度作了探讨。
【Abstract】 The experiments of culturing the explants of the leaves,stems,petals and ovaries of chrysanthemum were carried out in this paper. The results showed that different varieties and explants had different responses to the same medium. When the leaves were used as explants,the optimum media for Rm27-2 were MS+BA 0.5 mg/L+NAA 0.5 mg/L and MS+BA 2.0 mg/L+NAA 0.2 mg/L,the one for Rm 6-4 was MS+BA 6.0 mg/L+NAA 0.4 mg/L; the differentiation rate of Rm27-2 was higher than that of Rm6-4;the differentiation rate of stems was higher than that of leaves; the differentiation rate of the basal part of leaves was higher than that of the upper part of leaves;the optimum media of ovary culture were MS+BA 1.0 mg/L+NAA 1.0 mg/L and MS+BA 2.0 mg/L+NAA 2.0 mg/L;the medium MS+BA 2.0 mg/L+NAA 0.2 mg/L could effectively induce the petals to form calli and to differentiate. The function and proper concentration of cytokinin and auxin in the process of regeneration of explants were also discussed in this paper.
