嘉定白蒜茎尖脱毒技术研究

【摘要】 以嘉定白蒜为试材 ,采用茎尖分生组织培养技术获得无病毒植株。结果表明 :以带有一片叶原基、大小为 0 .2~ 0 .5mm的茎尖外植体 ,接种在附加BA0 .1mg L和IBA 0 .1mg L的MS培养基上 ,成苗率达 90 % ;不定芽增殖培养基以MS添加 6 BA0 .5mg L和IAA 0 .2mg L效果最好 ;筛选出生根最适培养基为MS +NAA0 .8mg L ,生根率达 70 %以上 ,组培苗经电镜检测脱毒率为 85% ,初步建立起脱毒嘉定白蒜繁殖体系。

【Abstract】 Virus-free plants of garlic were obtained by shoot tip culture of local garlic cultivated in Jiading. The results showed that when the medium was MS supplemented with 6-BA0.1mg/L and IBA0.1mg/L, and the size of shoot tip was 0.2~0.6mm with first leaf primordium for explant, the rate of plantlet developed was 90%; increase of regenerated buds was the most when the medium was MS containing 6-BA 0.5mg/L and IAA0.2mg/L; root formation medium that was MS added NAA0.8mg/L has been selected, and that ratio of the induction of root was more than 70%, regenerative plantlet rate of virus-elimination was 85% under electron microscope, and established preliminarily propagational system of virus-free garlic in Jiading.
