【摘要】 目的 用流式细胞仪测定高血压病人服用丹芎通脉颗粒前后血小板活化水平。方法 在服药前后,采用双注射器采集外周静脉血,3 8％枸掾酸钠抗凝,加入三种荧光素标记的单克隆抗体。分别加入FITC-PACl,PE-CD62P及PerCP-CD61单抗,室温避光孵育,1％多聚甲醛固定,上机检测。 结果 16例病人服药前后PACl+％从(12.61±3.15)％下降到(2.83±0.71)％(P<0.0001)。34例病人服药前后CD62P+％从(5.49±3.64)％下降到(2.31±1.54)％(P<0.0001)。结论 服用丹芎通脉颗粒后,病人的血小板活化水平都有显著性下降。更多还原

【Abstract】 Objective To determinate platelet activation in hypertensive patients treated with Danxiong Tongmai Keli by flow cytometry. Methods Before and after treatment, blood was taken by venipuncture without tourniquent with the first two milliliters of blood discarded, and 1 mL sample was collected, in 0. 1 mL of 3.8% sodium citrate. Within 3 min the 5 μL sample of FlTC-PAC, PE-CD62p and PerCP-CD61 were added to each 1 mL sample. After carefully mixing and incubating for 20 min at ambient temperature, samples were fixed in paraformaldehyde (1% ) for 10 min. All the specimens were assayed with flow-cytometry. Results PACl+ in 10 normals were (5.2 ±1.68)% and CD62P+ in 20 normals were (3.00 ±2.4)%. After treatment, PACl+ in 16 patients were decreased from ( 12.61 ±3. 15)% to (2. 83±0. 71 )% ( P <0.01 ) , CD62P+ in 34 patients were decreased from (5. 49±3. 64 ) % to (2.31±1. 54 )%(P< 0. 01 ) . Conclusion The platelet activation of hypertensive patients were declined significantly after treatment with Danxiong Tongmai Keli.更多还原
