绝经期糖尿病骨质疏松大鼠模型的建立及评价

【摘要】 目的:建立绝经期糖尿病骨质疏松症大鼠模型。方法:采用去卵巢与链脲佐菌素(STZ)注射复合法,观察大鼠骨代谢、骨形态、骨力学改变。结果:成模期大鼠体重下降,糖化血红蛋白(GHb)、骨钙素(BGP)、尿钙(Ca)/尿肌酐(Cr)水平上升,骨小梁平均厚度变小,最大凹入应力降低;成模后期骨代谢改变继续保持原态势,且尿脱氧砒啶啉(Dpd)与尿肌酐(Cr)、碱性磷酸酶(BALP)水平升高,降钙素(CT)、雌激素(E2)水平下降,骨小梁面积、最大压缩力、最大凹入应力降低。同步观察显示,假术组随着月龄增加,骨小梁平均厚度、骨小梁面积降低,但最大压缩力、最大凹入应力提高。结论:去卵巢糖尿病大鼠具有低体重、低雌激素、高糖化血红蛋白、骨力度下降、骨小梁减少、骨小梁间隔增宽等特点,骨松质骨丢失远大于密质骨等,且模型稳定性好,是研究绝经期糖尿病骨质疏松症较好的载体。

【Abstract】 Objective:To construct a rat model of postmenopausal osteoporosis diabetes.Methods:60 female SD rats aged 6 months were performed ovariectomy and injected streptozotocin to induce postmenopausal osteoporosis diabetes.And then bone metabolism,bone morphometriological and bone biomechanics were observed.Results:Rats in the model group showed decreased body weigh,increased glycosylated hemoglobin (GHb)level,BGP level and Ca/Cr level.The average thickness of bone trabecular and maximum depressing force(MDF) in the model group were decreased; In the late period,the change of bone metabolism in the model group kept the same level as before. Further more,in the model group the level of uric Dpd,uric Cr,and BALP increased,CT and E2 decreased.Area of bone trabecular,maximum compressing force(MCF),and MDF in the model group decreased.At the same time,with months passed by,sham operate group showed decreasement in average thickness and area of bone trabecular,increasement in MCF and in MDF.Conclusions:Postmenopausal osteoporosis diabetes rat model was stabile and good to use in studying postmenopausal osteoporosis of diabetes mellutus.
