钼-溴邻苯三酚红金属离子染料光谱探针测定尿蛋白
Determination of Protein in Urine Using Spectrum Probe Made up of Metal Ion Dye of Mo-Br-Pyrogallol Red
【摘要】 研究了在pH=2.5的NaAc-HCl缓冲溶液和吐温 80的介质中,钼(Ⅵ) 溴邻苯三酚红(BPR)形成的金属离子染料光谱探针,对白蛋白的显色体系,最大吸收峰为605nm,比试剂本身红移了21nm.在45~50℃水浴中加热15min,吸光度值可在1h内保持不变.线性范围为0 003~0 05(mg/mL)蛋白质,配合物的摩尔吸光系数为ε605=1.129×106(L·mol-1·cm-1), Sandelle灵敏度S=0 058(μg·cm-2).通过对尿液进行直接测定,其样品回收率为96 7%~100 4%,RSD=2 53%,结果令人满意.
【Abstract】 Metal ion dye of molybdenum and bromo-pyrogallol red (BPR) used as spectrum probe forms a coloring system with protein. The medium and pH of coloring system are studied . The results show that the maximum absorption of the coloring system is at 605nm with 21 nm of red shift compared to BPR itself in the medium of Tween 80 and pH 2.5 buffer. The system keeps steady in an hour after it’s heated for 15 min in 45~50℃ water bathing. The linear range is 0.003~0.05 mg/mL for protein .The molar absorptivity is 1.129×106Lmol(-1)cm(-1)and Sandelle sensitivity is 0.058ug·cm-2. The method has been applied for the determination of protein in the urine directly, with satisfactory. The RSD is 2.53% and the recovery is 96.7~100.4%.
