明胶/白芨胶多孔材料的细胞相容性研究

【摘要】 目的以明胶、白芨胶及中药黄连、丹参的提取物为原料,制备一种海绵状多孔材料,研究其与细胞的相容性熏为该产品作为创面敷料修复材料和皮肤、肌腱组织工程支架材料提供实验依据。方法将中药提取液复合到明胶和白芨胶支架材料中,通过冷冻干燥法制备成海绵状多孔材料熏以L-929细胞作为对象熏利用浸提液培养及材料表面直接培养法研究该材料的细胞相容性。结果载药材料在第2、4、7天的细胞相对增殖度在100.9%~111.3%范围内,其细胞毒性级别为0级;通过L-929细胞与明胶/白芨胶载药材料复合培养第2、7天的扫描电镜观察,L-929细胞在该材料上能很好地贴附、生长。结论明胶/白芨胶载药多孔材料有良好的细胞相容性,是可以安全使用的创面修复和皮肤、肌腱组织工程用材料。

【Abstract】 Objectives A new biomaterial made of gelatin, bletilla hyacinthine gum (BHG) and traditional Chinese herbs extract solution was prepared and its cytocompatibility was evaluated. Methods The Chinese herb extract solution was compounded with scaffolds (gelatin and BHG). Then it was frozen and dried to form spongy and porous material. The mouse fibroblast L-929 was cultured in soaking fluid of this biomaterial and on the material directly. Results The cells grew well in the extract solution of this biomaterial. The relative growth rate values in the 2nd, 4th and 7th day were within the scope of 100.9 %~111.3 %,the cell toxicity gradation is 0. On the 2nd and 7th day, a large number of L-929 cells adhering to and growing on the material were found under scanning electron microscope. Conclusion The biomaterial carrying Chinese herb prepared in this study has good cytocompatibility, and it is a safe biomaterial for application in wound healing and skin, tendon tissue engineering.
