低能陡脉冲致人卵巢癌SKOV-3细胞核DNA链断裂损伤的实验研究

【摘要】 目的 :研究低能陡脉冲对肿瘤细胞核DNA的损伤及诱导凋亡作用 ,分析其可能的机制及对电化学治疗潜在的影响。方法 :观察频率、脉宽、电压峰值固定 ,上升时间递增的低能陡脉冲对人卵巢癌细胞SKOV 3的损伤效应 ,用彗星试验检测细胞核DNA的损伤 ,计算拖尾率及拖尾长度以评估DNA损伤的程度 ,联合DNA片段化分析检测染色体断裂情况及电镜观察超微结构的改变。结果 :细胞核DNA拖尾率及拖尾长度与外加脉冲的上升时间有一定的量 效关系 ,DNA损伤、片断化程度随外加脉冲上升时间的增加而加重 ,电镜下出现典型的凋亡样改变。结论 :低能陡脉冲对瘤细胞核DNA有确切的损伤效应 ,可诱导瘤细胞凋亡。

【Abstract】 Abstract Objective:The aim of our study was to determine the damage effects of low energy steep pulse (LESP) on nuclei DNA of human ovarian cancer cell line,SKOV3 cell lines,to analysis its possible mechanisms and implications for electrochemotherapy.Methods:LESP with fixed frequency,width,peak value of voltage and gradually increased rise time of LESP were applied to SKOV3 cells.Comet assay was used to detect DNA damage of SKOV3,rate of tailedDNA and taillength of DNA migration were calculated for evaluation the degree of DNA damage,combination with DNA fragmentation analysis examined chromosomes breaks,and electron microscopy was used to detect ultrastructural changes.Results:A certain doseeffect relationship was observed between rate of tailedDNA,taillength and rise time of LESP.The degree of DNA damage and fragmentation positively correlated with rise time of LESP.A typical apoptosis also occurred under electron microscope.Conclusions:Our results showed that LESP has a certain damage effect on nuclei DNA of SKOV3 cell lines,also can induce apoptosis in these cells.
