哮喘患者树突状细胞分泌细胞因子的变化及其对Th1/Th2细胞因子的影响

【摘要】 目的 观察过敏性哮喘 (哮喘 )患者外周血单个核细胞 (PBMC)来源树突状细胞 (DC)分泌 IL- 12和IL - 10的情况 ,及其对 Th1 /Th2 型细胞因子平衡的影响。方法 分别取 9例哮喘患者和 14例正常人外周血 PBMC来源的 DC经 L PS刺激使其发育为成熟 DC(m DC)。另取脐血初始 T淋巴细胞 (Th0 )。Th0 和两组 m DC共培养 ,通过EL ISA法检测 m DC分泌的 IL- 12和 IL- 10及辅助性 T淋巴细胞 (Th)分泌的 IFN- γ和 IL- 4的含量。结果  1哮喘组 DC产生 IL - 12及其亚单位 IL - 12 p4 0和 IL - 10较对照组显著降低 (P <0 .0 1、 P <0 .0 5 )。2哮喘组 Th释放 Th1 型细胞因子 IFN- γ较对照组减少 (P <0 .0 5 ) ;Th2 型细胞因子 IL- 4较对照组显著增多 (P <0 .0 1)。3两组 IL- 12和 IFN- γ均呈正相关 (r =0 .75 81、 P <0 .0 1,r =0 .6 0 2 8、 P <0 .0 5 ) ,IL - 12和 IL - 4均呈负相关 (r =- 0 .74 98、 P <0 .0 1,r=-0 .74 81、 P <0 .0 1)。4哮喘组 IL- 10与 IFN- γ呈正相关 (r =0 .6 4 37,P <0 .0 5 )。5两组 IL- 12和 IL- 10均呈正相关 (P均<0 .0 1)。结论 哮喘患者 DC分泌 IL- 12和 IL- 10减少 ,导致 Th0 向 Th2 优势分化 ,细胞因子平衡向 Th2 型倾斜 ,合成 IL-4增多和 IFN-γ减少 ,后者是哮喘发生的重要机制

【Abstract】 Objective To observe interleukin-12(IL-12) and interleukin-10(IL-10) produced by human peripherial blood monocyte(PBMC)-derived DC in allergic asthmatic subjects,and its influence on Th 1- and Th 2-cytokine pattern balance.Methods PBMC-derived immature DC(iDC) were obtained from peripherial blood samples in 9 asthmatic patients and in 14 healthy volunteers.iDC were stimulated with LPS as mature DC(mDC).Naive T cells(Th 0) were sorted from umbilical cord blood.Th 0 anh mDC from two groups were cocultured in complete RPMI1640 media respectively.IL-12 and IL-10 released by mDC and interleukin-4(IL-4) and IFN-γ produced by Thelpercells(Th) were measured by ELISA.Results The level of IL-12,IL-12p40 and IL-10 produced by DCs in asthmatic subjects group was all significantly lower than that in healthy control group(P<0.01,P<0.05 respectively).IL-4 production by Th 2 which were primed by DCs from those cases with asthma was significantly increased than that from control group(P<0.01),contrarily,IFN-γ in patients group was reduced than that in control group(P<0.05).The level of IL-12 was positively correlated to IFN-γ in two groups(P<0.01,P<0.05 respectively),and negatively correlated to IL-4 in two groups(all P<0.01).IL-10 was posively correlated to IFN-γ in patients group(P<0.05).There was positive correlation between IL-12 and IL-10 in two groups (all P<0.01).Conclusion Because of diminished IL-12 and IL-10 production,DC preferentially induce Th 2 cell development which bias Th 1-and Th 2-cytokine pattern balance towards Th 2-cytokine pattern resulting in increased IL-4 production and reduced IFN-γ production by TCs,which is one of important pathogensy for asthma.
