【摘要】 目的　探讨人胎儿卵泡形成过程中卵细胞凋亡及其凋亡蛋白 Caspase- 3的表达意义。方法　收集13例行水囊引产终止妊娠的胎儿卵巢 ,其中孕中期 (2 0～ 2 8孕周 ) 7例 ,孕晚期 (2 9～ 38孕周 ) 6例。利用原位末端标记法 (TUNEL法 )和免疫组化法观察胎儿卵泡形成过程中卵细胞凋亡及其凋亡蛋白 Caspase- 3的表达。结果　部分胎儿卵细胞末端标记阳性 ,孕中期卵细胞凋亡指数 [(9.8± 1.2 ) % ]大于孕晚期 [(3.1± 1.0 ) % ],P<0 .0 1。凋亡相关蛋白 Caspase- 3在胎儿卵细胞内广泛表达 ,孕中期阳性单位 (36 .6± 3.1)大于孕晚期 (2 0 .2± 2 .6 ) ,P<0 .0 1。Caspase- 3表达与卵细胞凋亡呈正相关。结论　胎儿卵泡形成过程中卵细胞发生凋亡 ,导致生殖细胞丢失 ,凋亡蛋白 Caspase- 3可能参与卵细胞凋亡的调控更多还原

【Abstract】 Objective To investigate the oocyte apoptosis and the expression of apoptosis related protein Caspase 3 in the oocytes during human fetal follicular development Methods Ovarian tissues from 13 fetuses(aged 20 to 38 weeks) were obtained after abortion induced by insertion of water sac, including 7 fetuses of midpregnancy (aged 20 to 28 weeks) and 6 fetuses of late pregnancy ( aged 29 to 38 weeks) Using TUNEL method and immunohistochemistry, the occurrence of oocyte apoptosis and the expression of apoptosis related protein Caspase 3 during human fetal follicular development were examined Results Some oocytes were TUNEL positively stained The apoptotic index of oocytes of midpregnancy [(9 8±1 2)%]was higher than that of late pregnancy [(3 1±1 0)%],P<0 01 The expression of apoptosis related protein Caspase 3 was also noticed in the fetal oocytes, and its PU value of midpregnancy(36 6±3 1) was significantly higher than that of late pregnancy(20 2±2 6), P<0 01 There was a positive relationship between Caspase 3 expression and apoptosis in the oocytes Conclusion The oocyte apoptotosis occurs and causes germ cell loss during human fetal follicular development Apoptosis related protein Caspase 3 may participate in modulating the oocyte apoptosis更多还原
