Angiostatin基因对C6胶质瘤体内生长的抑制作用
Inhibitory efficiency of angiostatin cDNA on glioma growth in vivo
【摘要】 目的:研究血管生成抑制因子angiostatin对C6胶质瘤体内生长的影响。方法:以阳离子脂质体介导将angiostatin基因导入C6胶质瘤细胞,G418(1000μg/m1)克隆筛选、扩大培养后接种于裸小鼠皮下,观察移植瘤生长的体积、重量变化及肿瘤抑制率,应用SABC免疫组化技术检测肿瘤微血管密度。结果:angiostatincDNA对体内生长的胶质瘤有显著抑制作用,抑瘤率可达82.2%(P<0.01),转染组胶质瘤中微血管密度眼(6.8±2.1)/HP演低于对照组眼(16.7±4.8)/HP演(P<O.01)。结论:angiostatincDNA能通过抑制血管生成进而抑制胶质瘤生长,在胶质瘤临床治疗中有应用价值。
【Abstract】 Objective: To explore the inhibitory role of angiostatin cDNA on the growth of C6 glioma in vivo and its possibility to be a new anti-tumor drug. Methods: C6 glioma cells were transfected with angiostatin cDNA by a lipofectin method. Four weeks after transfection, G418(1 000 μg/m1) resistant cells were selected and subcutaneously implanted into nude mice. The tumor volume, weight and inhibition rates were observed. The glioma microvessel density was detected by SABC immunohistochemical technique. Results: Angiostatin cDNA inhibited the growth of C6 glioma remarkably, the tumor inhibition rate being 82.2%(P<0.01).
