降钙素在围植入期小鼠子宫内膜中的表达

【摘要】 目的:探讨降钙素在胚泡植入过程中的生物学作用。方法:取未孕动情期、真孕及假孕3~8d的小鼠子宫作切片,用免疫组织化学方法和图像分析方法对降钙素在围植入期子宫内膜中的表达进行定性和定量分析。结果:降钙素仅分布于子宫内膜腺上皮。在真孕及假孕组小鼠,受精后第3~4d(植入前),子宫内膜腺上皮中降钙素的含量均明显高于未孕组(P<0.01);受精后第6~7d(植入后),真孕小鼠子宫内膜中降钙素的含量进一步增多,明显高于假孕及植入前各时间组(P<0.01)。结论:①降钙素参与胚泡植入过程,还可能间接参与植入后胚泡的滋养层扩展、分化及子宫内膜蜕膜化反应;②降钙素在子宫内膜中的表达不仅受母体因素调控,还与胚泡的刺激有关。

【Abstract】 Objective:To study the biologicale effects of calcitonin in the process of blastocystimplantation.Methods:The uteri of nonpregnant(estrous stage)and day 3~8 of pregnant,pseudopreg-nant mice were sectioned.The distribution and changes of calcitonin in the peri-implantation mouseendometrium were examined by immunohistochemistry.An image analysis was also performed.Results:The calcitonin-specific staining was only localized in the endometrial glandular epithelial cells.In preg-nant and pseudopregnant group,day 3~4(post fertilization)of gestation,the level of calcitonin was sig-nificantly enhanced compared with that in the nonpregnant group(P<0.01); Day 6-7 of gestation,the ex-pression of calcitonin increased dramatically compared with those in the corresponding stagage of pseudo-pregnancy and in pre-implantation period of pregnancy(P<0.01).Conclusion:①Calcitonin,in addition tocorrelating to blastocyst implantation,may indirectly participate in the process of trophoblast outgrowth,differentiation and decidualization of endometrium.②The expression of calcitonin in mouse endometriumis regulated not only by maternal steroid hormones,but also by the implanted embryos.
