杜仲树皮cDNA文库的构建与分析
Construction and analysis of a cDNA library in Eucommia ulmoides Olive
【摘要】 以杜仲EucommiaulmoidesOlive五六月份的幼嫩树皮为材料,提取总RNA,采用RT-PCR技术(逆转录—聚合酶链式反应)合成cDNA双链,以Uni-ZAPXRvector为载体,构建了杜仲树皮的cDNA文库,其文库的滴度为3.4×1010pfu·mL-1,含插入片段的频率是99%,插入片段的分子长为400~2500bp,为进一步研究杜种特有的药用成分合成途径及杜仲胶合成途径的关键酶基因奠定了基础。
【Abstract】 The total RNA from the new barks of Eucommia ulmoides Olive in May or June were extracted,A cDNA library were constructed from Eucommia ulmoides Olive barks by RT-PCR techniques(reverse transcription polymerase chain reaction)and Uni-ZAP XR vector.The library consisted of 3.4×1010pfu·mL-1,in which 99% of the phages were recombinant.Insert sizes ranging from 400bp to 2 500bp were obtained,which would found the base to further study the key enzymes of the biosynthetic pathway of medical constituents and the rubber in Eucommia ulmoides Olive.
