单细胞凝胶电泳评价顺铂致人卵巢癌细胞DNA损伤修复的实验研究
Study on the DNA damage repair induced by cisplatin in human ovarian cancer cell line HO8910 using comet assay.
【摘要】 目的 采用单细胞凝胶电泳 (彗星实验 )研究顺铂致人类卵巢癌HO8910细胞DNA的损伤及修复 ,为彗星实验应用于临床检测肿瘤细胞对化疗药物的敏感性奠定基础。方法 细胞采用 0 0 2EC5 0、0 0 3EC5 0及 0 0 5EC5 0三种浓度顺铂处理 1h ,更换培养基后 ,继续培养 3h、2 4h和 48h进行彗星电泳实验。结果 顺铂处理后造成HO8910细胞产生明显的DNA损伤 ,呈现不同彗尾长度的彗星。顺铂浓度增加 ,平均彗尾长度增加 ;一定顺铂浓度下 ,彗尾长度随培养时间的延长逐渐减小 ,较低浓度 ( 0 0 2EC5 0 )处理的细胞在培养 48h后彗尾长度可恢复至接近对照组的水平 ;在顺铂处理0 0 2EC5 0浓度下随培养时间延长 ,彗星出现比率逐渐下降 ,而 0 0 5EC5 0浓度处理的细胞彗星比率则呈上升趋势。结论彗星实验检测DNA损伤及修复的敏感度较高 ,H0 8910细胞可修复一定浓度的顺铂引起的DNA损伤
【Abstract】 Objective To study the DNA damage and repair induced by cisplatin in human ovarian cancer cell line HO8910 using comet assay.Methods The cells were treated with three concentrations of cisplatin as 0.02 EC50,0.03 EC50 and 0.05 EC50 respectively for one hour at 37℃,followed by a continued incubation for 3h,24h and 48h,than the comet assays were performed to observe the tail length.Results Cisplatin of different concentrations induced significant comet in HO8910 cells,the average tail length increased as the cisplatin concentration raised.The tail length decreased as the incubation time prolonged under the given cisplatin concentration,the cells treated with lower concentration(0.02 EC50)could recover nearly to the level of control after 48 h incubation.The percentages of comet reduced as the incubation time prolonged under the 0.02 EC50 concentration but increased for the 0.05 EC50 concentration.Conclusion HO8910 cells can repair the DNA damage induced by cisplatin of certain concentrations,and the comet assay had considerable sensitivity to detect the lesion and repair process,could be used to estimate the sensitivity of tumor cells to the chemotherapy drugs in clinical test.
