生物功能化色谱新概念及其方法学研究

【摘要】 生物体内大多数生物过程在某种意义上与色谱的作用机理类似, 因此我们提出一个色谱新概念,即利用仿生固定相和流动相模拟生物过程。目前色谱的应用主要局限在它的分离和定量功能。如果使固定相具有生物特性,即利用色谱模拟生物过程,将不仅进一步拓展色谱的应用空间,而且也可以解决大量的生命科学中的问题。生物功能化色谱尚未见报道,这个新技术不仅可以迅速、准确地测定生物特性,同时结合色谱的分离能力可以实现高通量测定。在本研究中我们发展了用于模拟多巴胺代谢的生物功能化色谱。根据影响多巴胺在体内代谢过程的相关酶以及它们的作用机理,通过构筑具有生物活性的色谱固定相和流动相把生物分子之间的作用过程用色谱模拟出来。这个具有生物功能的色谱已经尝试性地用于帕金森病的药物筛选。
【Abstract】 Most of the biological processes are somehow similar to the mechanism of chromatography, and thus a new concept wasproposed that the chromatography could be applied to simulate the biological process by means of the bionic stationary and mobilephases. The new technique, namely the biologically functionzlized chromatography (BioFC), not only is able to measure biologicalproperties, such as the protein-protein interaction, but also enable a high throughput because of the powerful separation ability offeredby the chromatographic mechanism. In this article, a biological functionzlized chromatographic system was designed for imitating themetabolism of dopamine in the brain. Based on enzymes and proteins involved in the metabolism of dopamine, the BioFC system wasbuilt as follows: the membrane proteins were dynamically fixed on the immobilized artificial membrane which is used as the stationaryphase, and the cytosol proteins and molecules were in the mobile phase. The BioFC was preliminarily applied to the drug screening ofParkinson’s disease.
