聚醚酯材料的生物相容性评价和水解降解实验研究

【摘要】 以大鼠血管成纤维细胞为模板细胞,按照ISO10993标准,测试了聚醚酯材料聚对苯二甲酸丁二醇酯-co-聚环己烷二甲酸丁二醇酯-b-聚乙二醇嵌段共聚物(DA)的细胞毒性。采用倒置显微镜观察细胞在DA膜片上的粘附和生长情况,并用MTT法测定细胞增殖率。采用气泡法测定DA薄膜的接触角。在37℃下,pH=7.4的磷酸盐缓冲溶液中研究水解降解反应。结果表明:DA系列聚醚酯材料无严重细胞毒性;成纤维细胞在DA薄膜表面上贴附生长良好;材料亲水性好,其重均分子量在水解过程中随时间的延长逐渐降低,材料发生明显降解。DA有作为生物医用材料的可行性。

【Abstract】 Mouse fibroblasts were used as model cells to test the cytotoxicity of poly(butylene terephthalate)-co-poly(butylene cyclo-hexane dicarboxylate)-b-poly(ethylene glycol) (DA) multiblock copolymers according to ISO10993 standard. The morphology of cell adhesion on DA films was observed and photographed by invert light microscope. Using MTT method tested the cell growth rate. Contact angles of DA. copolymers were determined using the captive bubble technique. Hydrolytic degradation experiments on DA films were carried out in phosphate buffer saline (PBS) of pH 7.4 at 37℃. The extraction of DA showed no acute in vitro cytotoxicity. Mouse fibroblasts adhere to the DA films and grow well. The weight molecular weight (Mw) of DA copolymers decreased obviously with the degradation time. DA has the potential to be used as biomaterial.
