蜗牛酶水解牛蒡子苷制备苷元的研究
Preparation of Arctigenin by Enzymolysis of Arctiin with Snail Hydrolase
【摘要】 [目的]优选牛蒡子苷的酶解3-艺条件。[方法]用蜗牛酶对牛蒡子苷进行酶解,采用正交设计优选酶解工艺条件,高效液相色谱法测定酶解产物牛蒡子苷元的含量。[结果]在37℃条件下用1/5底物量的蜗牛酶,于pH 5的无菌水或生理盐水中酶解36~48 h最为适宜,确定了含醇水溶性有机溶剂作为酶解终止制。[结论]在所确定的条件下牛蒡子苷的酶解得率达72%~78%,酶解终止剂选择得当,工艺便于放大;检测方法简便、准确。
【Abstract】 [ Objective ] To optimize the condition for enzymolysis of arctiin by snail hydrolase. [ Methods ] Snail hydrolase was used for the enzymolysis of arctiin and the process condition of enzymolysis was optimized by orthogonal design. Arctigenin content was measured by high performance liquid chromatography (HPLC) . [Results] The optimum hydrolysable conditions were achieved at a pH 5 sterilized solution of water or normal saline for 36 ~ 48 hours of enzymolysis with snail hydrolase which was 1/5 amount of the substrate arctiin. Water-soluble organic solution containing alcohol was a suitable terminator of enzymolysis. [Conclusion] The yield rate of arctigenin is up to 72% -78% under the optimized conditions; the process can be applied for a larger production when a suitable terminator is used. Therefore, quantitative assay for arctigenin by HPLC is simple and accurate.
