32P标记白细胞在异种角膜移植中的应用
Application of 32P-labeled White Cells in Xenocorneal Transplantation
【摘要】 目的通过32P标记供体白细胞预处理受体,观察含β射线的32P在异种(鸡→兔)角膜移植中的抗免疫排斥作用。方法建立异种(鸡→兔)角膜移植实验模型,术前2周将32P标记供体白细胞注入兔血管内预处理受体,术后观察新生血管长至植床植片交界处的时间,植片透明、半透明和完全混浊的时间,同时检测移植家兔的外周血白细胞、T淋巴细胞在术前、术后的变化。结果应用32P标记供体白细胞预处理受体组角膜移植后,其存活时间与其它三组相比明显延长(P<0.01),其外周血T淋巴细胞降低,表明该方法处理角膜具有一定抑制淋巴细胞分裂增殖的作用。术后2周血白细胞数量无明显变化,表明小剂量的32P静脉注射是安全的。结论32P标记供体白细胞预处理受体为抗角膜移植免疫排斥反应提供了新的手段。
【Abstract】 Objective To investigate the immnosuppression of rejection response of chicken to rabbit xenocorneal transplantation after pretreatment of recipient by 32P labeled white cells. Methods An experimental model of chicken to rabbit xenocorneal transplantation was established. The 32P labeled white cells were implanted into the vasocular of rabbit, to pretreat this recipient for two months before operation. The new bored vasocular and the transparence of transplanted cornea were observed. The amounts of white cells and T lymphocytes were compared before and after operation. Results The survival of transplanted cornea was significantly prolonged(P< 0.01), and T lymphocytes slightly decreased after the cornea was pretreated by 32P labeled white cells compared with the other three groups, indicating that the pretreatment of cornea inhibited the proliferation of lymphocytes. The white cells did not show obvious changes among the four groups two weeks after operation, showing the safety of low dose treatment of 32P labeled white cells. Conclusion The pretreatment of recipient by 32P labeled white cell was safe and could inhibit the proliferation of lymphocytes in the transplanted cornea, which might become a hopeful alternative in immnosuppression of rejection in cornea transplantation.
