【摘要】 目的研究日本血吸虫成虫血红素加氧酶-1(HO-1)基因的表达并对部分编码区序列进行测序分析。方法收集日本血吸虫成虫,提取总RNA;以成虫总RNA为模板,利用HO-1特异性引物一步法RT-PCR进行体外扩增,检测血吸虫成虫是否有HO-1基因的转录表达;纯化PCR扩增产物,DNA测序后进行序列分析。结果提取较高纯度的血吸虫成虫总RNA后,RT-PCR扩增得到一特异性条带,测序结果显示该特异性片段长度为506bp。结论本研究用RT-PCR的方法证实了HO-1基因在血吸虫体内的转录表达,为进一步研究血吸虫HO-1基因的全长和基因调控奠定了基础。更多还原

【Abstract】 Objective To explore the expression of HO 1 transcript of Schistosoma japonicum and to sequence the partial cDNA. Methods Adult worms of S.japonicum were collected and total RNA was isolated. The expression of HO 1 cDNA was detected by RT PCR. Sequence analysis was carried out after purification of the amplified product. Results High quality RNA was isolated from adult worms. One specific band was amplified from cDNA by RT PCR. Sequencing result revealed the band was 506bp. Conclusion Expression and of HO 1 gene in adult worms of S.japonicum was demonstrated by RT PCR.更多还原
