三氧化二砷对人白血病细胞株NB4和K562及MOLt4的增殖、周期及凋亡的影响

【摘要】 目的 :观察三氧化二砷 (As2 O3 )对人白血病细胞株NB4、K5 62、MOLt4的增殖 ,周期及凋亡的影响。方法 :采用台盼蓝拒染法计数活细胞数 ,并计算细胞生长率 ;细胞涂片经Wright Giemsa染色 ,光镜下观察细胞的形态 ;流式细胞仪解析细胞周期。结果 :1)As2 O3 能够抑制NB4、K5 62、MOLt4细胞的增殖。 2 ) 4μmol/LAs2 O3 能够使K5 62细胞发生M期阻滞。但NB4、MOLt4细胞未见明显改变。 3 ) 2、4μmol/L的As2 O3 处理NB4、MOLt4和K5 62细胞 48h ,NB4、MOLt4细胞株的凋亡细胞明显增多。但是 ,K5 62细胞未见明显改变。结论 :As2 O3 不但对NB4细胞具有抑制增殖 ,诱导细胞凋亡的作用 ,而且对非t( 15 ;17)的K5 62、MOLt4细胞也具有抑制增殖的作用 ,同时诱导MOLt4细胞凋亡 ,使K5 62细胞发生M期阻滞。

【Abstract】 OBJECTIVE:To investigate the impact of arsenic trioxide (As 2O 3) on proliferation, cell cycle and apoptosis in human leukemia cell lines NB4, K562 and MOLt 4.METHODS: Cell viability and cell growth rate were determined by the Trypan blue exclusion. Cell morphology was observed under the light microscope with Wright-Giemsa staining. Cell cycle was analysed by the flow cytometry.RESULTS: 1)As 2O 3 inhibited the proliferation of NB4, K562 and MOLt4 cells. 2) As 2O 3 at the concentration of 4 μmol/L induced M phrase arrest of K562 cells,but no significant changes were observed in NB4 and MOLt 4 cells. 3) After treated with 2 μmol/L and 4 μmol/L As 2O 3 for 48 hours,the percentage of apoptosis markedly increased in NB4 and MOLt4 cell,but no change was observed in K562 cells under the same condition. CONCLUSIONS: As 2O 3 has the effect of growth inhibition and apoptosis induction in NB4 cells and can inhibit the proliferation of K562 and MOLt4 cells without t (15;17). It can also induce apoptosis in MOLt4 cells and block K562 cells at M phase.
