【摘要】 目的 :研究环氧合酶 -2 (Cy clooxygenase 2 ,COX 2 )在子宫内膜癌及癌前病变组织中的表达 ,以探讨其与子宫内膜癌发生、发展的关系。方法 :应用逆转录聚合酶链反应 (RT PCR )技术 ,检测 10例正常子宫内膜 ,10例不典型增生子宫内膜 ,3 2例子宫内膜癌组织中COX 2mRNA的表达。结果 :COX 2mRNA在正常子宫内膜、不典型增生子宫内膜及子宫内膜癌中的表达率分别为 2 0 0 %、70 0 %和 90 6% ,相对含量分别为 5 0 87、79 5 8± 8 0 9和 91 73±3 80。子宫内膜癌及不典型增生子宫内膜中COX 2mRNA的表达率和相对含量明显高于正常子宫内膜 ,差异有统计学意义 ,P <0 0 0 1;不典型增生子宫内膜与子宫内膜癌COX 2mRNA的表达率比较 ,差异无统计学意义 ,P >0 0 5 ;但子宫内膜癌组织中COX 2mRNA的相对含量明显高于不典型增生子宫内膜 ,P <0 0 1。在子宫内膜癌组织中 ,中、低分化及有深肌层浸润组COX 2mRNA的相对含量明显高于高分化及浅肌层浸润组 ,P <0 0 1。结论 :COX 2在子宫内膜癌的发生、发展中起重要作用 ,并与子宫内膜癌的某些生物学行为密切相关更多还原

【Abstract】 OBJECTIVE:To investigate the relationship between the expression of mRNA and carcinogenesis and development of the endometrial carcinoma.METHODS:Quantitive reverse transcription-polymerase chain reaction (RT-PCR) was used to examine the levels of COX-2 mRNA in the following groups:10 cases with normal endometrium,10 cases with hyperplasia endometrium and 32 cases with endometrial carcinoma.RESULTS:The positive percentages of COX-2 mRNA expression in normal endometrium,hyperplasia endometrium and endometrial carcinoma were 20.0%,70.0% and 90.6%,respectively,and the positive rates in endometrial carcinoma and hyperplasia were singnificantly higher than that in normal endometrium,P<0.001,but there was no statistical significance between hyperplasia and endometrium carcinoma,P>0.05.The relative content of COX-2 mRNA was 50.87,79.58±8.09 and 91.73±3.80,respectively,and there were singnificantly differences among these groups,P<0.01.The relative content of COX-2 mRNA in moderately and poorly differentiated tissues of endometrium carcinoma was greater significantly than that in well differentiated tissues,P<0.01.Similarly,the relative content of COX-2 mRNA in endometrial carcinoma with deep myometrial invasion was higher significantly than that without or less than 50% myometrial invasion,P<0.01.CONCLUSION:COX-2 may play an important role in the carcinogenesis and development of endometrial carcinoma,and correlates with the biological behavior of endometrial carcinoma.更多还原
