Bim与p-Bad蛋白在子宫内膜癌中的表达及意义
Expressions of Bim and phosphor-Bad in endometrial carcinoma
【摘要】 目的 :探讨仅含BH3区域蛋白 (BH 3 onlyproteins)家族中Bim与 p Bad蛋白在子宫内膜癌中的表达及其意义。方法 :采用免疫组织化学方法和免疫印迹法 (Westernblot)对 5 5例子宫内膜癌、13例子宫内膜非典型增生组织中的Bim及 p Bad蛋白表达进行检测 ,并以 11例正常子宫内膜组织作对照组。结果 :Bim蛋白阳性表达在对照组最多 (x- =1 67) ,在子宫内膜非典型增生 (x- =1 2 9)及癌组织中 (x- =0 71)明显减少 ,3组比较差异有统计学意义 ,F =10 82 9,P <0 0 1;p Bad蛋白阳性表达在对照组中表达量最低 (x- =0 11) ,在子宫内膜非典型增生 (x- =1 2 4)及癌组织中 (x- =2 0 5 )明显增加 ,3组比较差异有统计学意义 ,F =12 44 4,P <0 0 1。在高中分化腺癌中 ,p Bad表达显著增加 (x- =2 2 8) ,与对照组相比 ,差异有统计学意义 ,P <0 0 1。Bim和 p Bad的表达在子宫内膜癌各临床分期及肌层浸润之间的差异均无统计学意义。结论 :Bim表达减少可能是子宫内膜癌发生过程中的早期事件 ;Bad促凋亡活性减弱是导致子宫内膜癌发生的原因之一 ;提示仅含BH3区域蛋白家族中Bim、Bad与子宫内膜癌的发生关系密切
【Abstract】 OBJECTIVE:To investigate the role of members of BH3-only proteins Bim and phosphor-Bad in the occurrence of endometrial carcinoma.METHODS:The expressions of Bim and phosphor-Bad in endometrial carcinoma and endometrial atypical hyperplasia were detected by immunohistochemistry and Western blot,and the normal endometrium was adopted as the control.RESULTS:Expression of Bim in normal endometrium (x-=1.67) was higher than those in atypical hyperplasia (x-=1.29) and carcinoma (x-=0.71).The difference in the three group was significant,F=10.829,P<0.01. On the contrary, p-Bad expression in normal endometrium (x-=0.11) was much lower than those in atypical hyperplasia (x-=1.24) and carcinoma (x-=2.05),F=12.444,P<0.01. Compared with normal endometrium, the expression of p-Bad in well and mid differentiated carcinoma was increased very significantly, P<0.01.However,among the different clinical stages and muscle invasion of endometrial carcinoma, the expressions of the two proteins were of no difference.CONCLUSIONS: The decrease of Bim may be the early affair during the development of endometrial carcinoma.Weaken of the pro-apoptotic activity of Bad is one of the causes of endometrial carcinoma. All of these suggest that there is close relationship between the BH3-only proteins Bim, Bad and endometrial carcinoma.
