石蜡包埋肺癌组织DNA提取方法的改进
DNA extraction from paraffin-embedded lung cancer tissue and PCR
【摘要】 目的 :探讨从保存肺癌标本中提取基因组DNA的改良方法。方法 :石蜡包埋肺癌组织在二甲苯脱蜡后 ,先用PBS缓冲液 (设立pH值梯度 ) 60℃温浴处理 4~ 8h ,接着用不同梯度乙醇处理 ,最后再用PBS缓冲液常温处理 4~ 8h。结果 :用PBS缓冲液预处理比直接用乙醇梯度处理所获得的DNA质量要高 ,且pH值在 7 5~ 8 5时最佳 ,PCR扩增产物长度可达 675bp左右。结论 :保存时间较长的标本用偏碱性 (pH值 7 5~ 8 5 )PBS缓冲液预处理 ,能提高其DNA质量
【Abstract】 OBJECTIVE:To extract the genomic DNA from preserved specimen and apply it for molecular research.METHODS:After being taken off paraffin wax with xylene from paraffin-embedded lung cancer tissue, first the tissue was infused into PBS buffer for 4-8 hrs in water bath, the temperature was 60℃, then handled with different concentration of alcohol.RESULTS:Long DNA fragment was acquired by this method, and higher yields of DNA were got at pH 7.5-8.5.The PCR products amounted to 675 bps.CONCLUSIONS:Long DNA fragment can be acquired from the PBS buffer (pH 7.5-8.5) pretreatment preserved specimen.It is important for molecular research of sample DNA.
