【摘要】 目的 :探索胰岛素样生长因子 -1(in sulin likegrowthfactor 1,IGF 1)及其受体 (in sulin likegrowthfactor 1receptor ,IGF 1R)在胃癌发生、发展中的作用 ,寻找新的胃癌早期诊断标志物 ,并为胃癌的病因研究提供理论依据。方法 :对 2 8例经手术切除并由病理证实的胃癌患者的癌、癌旁及胃黏膜正常区组织标本进行研究 ,以 14例正常人胃黏膜标本作对照 ,应用RT PCR方法 ,检测IGF 1/IGF 1R在胃癌组织与正常人胃黏膜中的表达变化和胃癌患者的癌、癌旁及胃黏膜正常区组织中的表达变化。结果 :IGF 1/IGF 1RmRNA的表达量在胃癌组明显高于正常对照组。在胃癌组织中IGF 1/β actin比值为 0 3 0 5± 0 0 3 2 ,IGF 1R/β actin比值为 0 12 9± 0 0 11;在正常胃黏膜中IGF 1/β actin比值为 0 15 4± 0 0 13 ,IGF 1R/β actin比值为 0 0 2 7± 0 0 0 4。两组间的差异有统计学意义 ,P <0 0 1;且mRNA的表达量按正常、癌旁、癌组织顺序逐渐增高 ,3组间差异有统计学意义 ,P <0 0 5。结论 :IGF 1/IGF 1R因子的高表达 ,促进了胃黏膜上皮细胞的异常增殖 ,在胃癌的发生发展中起重要的作用。更多还原

【Abstract】 OBJECTIVE: To study the expression of insulin like growth factor 1 and insulin like growth factor 1 receptor (IGF 1/IGF 1R) in gastric mucosa of patients with gastric carcinoma, so as to explore its mechanism and find an useful indicator for early diagnosis of gastric carcinoma. METHODS:The mRNA expression levels of IGF 1/IGF 1R were assessed by RT PCR method in the gastric cancer tissues, adjacent mucosa and tumor free tissues from 28 patients with gastric carcinoma and the normal gastric mucosa from 14 healthy controls. Tissue specimens were obtained by biopsy, and confirmed by histological evaluation. RESULTS:The mRNA levels of IGF 1/IGF 1R were increased in gastric cancer tissues compared with normal tissues from healthy controls, P <0 01 . The ratios of IGF 1/β actin and IGF 1R/β actin were 0 305±0 032 and 0 129 ±0 011 in gastric cancer tissues, but the ratios were 0 154±0 013 and 0 027 ±0 004 in the tumor free tissues. There was a significance between gastric cancer tissues and the tumor free tissues, P <0 01. The mRNA levels of IGF 1/IGF 1R increased from the tumor free tissues, adjacent mucosa to gastric cancer tissues, P <0 05 . CONCLUSIONS:The overexpression of IGF 1/IGF 1R is of great significance in the carcinogenesis and development of gastric carcinoma,and it might be used as an indicator for early diagnosis for gastric carcinoma.更多还原
