【摘要】 建立高效液相法(HPLC)测定肿瘤患者外周血血管抑素(Angiostatin)含量水平的方法。反相"C4 Wide Pore柱(4.6×210mm)为色谱柱,流动相A为0.1％TFA、90％乙酸缓冲液;流动相B为0.1％TFA,进行四段梯度洗脱。HPLC流速为1mL／min,温度26℃。荧光检测器测得血管抑素回收率为97％,回归方程线性良好。该方法简洁、快速,灵敏度高,重现性好,可有效分析人血清中血管抑素及其类似物的含量水平。更多还原

【Abstract】 Objective High - performance liquid chromatographic(HPLC) method for the determination of angiostatin in the serum in patients with cancer. The derivatives were separated on a C4 Wide Porecolumn(4.6 × 210mm) , and eluted with 0.1% TFA using a four - step linear gradient.The flow rate was 1m 1/min and the temperature of column was 261 .The column eluate was monitored by fluorescence detection. The recovery of angiostatin was 97% .The relative coefficiencts was above 0.99. Therefore, it’ s a simple,rapid and sensitive method that can be helpful in determining angiostatin in the serum with cancer patients.更多还原
