糖尿病合并感染大鼠胰岛细胞凋亡的变化

【摘要】 目的 探讨糖尿病合并感染动物模型胰岛细胞凋亡的变化。方法 盲肠结扎穿刺术(CLP)建立感染的动物模型,模型是在键佐菌脲菌素(STZ)诱发血糖升高的基础上行盲肠结扎穿刺术(CLP)术,对大鼠的胰岛细胞DNA进行琼脂糖凝胶电泳法测胰岛细胞凋亡,之后做胰岛细胞凋亡的定量分析。结果 正常组胰岛细胞DNA显基因组条带位于加样孔的附近,疾病组胰岛细胞DNA呈现为梯状条带。糖尿病合并感染组细胞凋亡率显著性升高(均P<0.05);糖尿病组的细胞凋亡率与感染组的差异也具有显著性(P<0.05)。结论 糖尿病和/或感染产生的复杂的诱导因子作用下发生胰岛细胞凋亡以及功能的改变,其中糖尿病合并感染的动物更为明显。

【Abstract】 Objective To investigate apoptosis of pancreatic islet cells in rats with diabetes mellitus complicated infection. Metheds Diabetic model of rats was induced by stregtozotocin(STZ) ; Infected animal model was established by ceccal ligation and puncture(CLP) ; Apoptosis of islet cells were detected by 2% agarose electrophoresis, then were measured by the semi - quntitative analysis. Results DNA from normal cell was inside sample - hole, and DNA of diseases animals were showed narrow strip shaped like a staircase. Ratio of apoptosis was elevated significantly in rats with DM complicated infection(P< 0.025) .The Ratio was increased in rats with DM compared to infected rats. Conclusions DM and/or infection lead to apoptosis of pancretic islet cells and injure the fuction of islet, specific in the rats with DM complicated infection.
