实验性脑出血大鼠脑组织NF-κB的表达及意义
The Expression and Significance of NF-κB in Rat Brain after Experimental Intracerebral Hemorrhage
【摘要】 目的 探讨大鼠实验性脑出血后脑组织NF -κB (nuclearfactor-kappaB)的表达规律及意义。 方法 成年雄性SD大鼠随机分为脑出血组 (4 2只 )和假手术组 (6只 )。脑出血组又分为 1h、 3h、 6h、 2 4h、 4 8h、 72h、7d 7个亚组 ,每亚组 6只。应用干 -湿重法观察脑水肿规律、HE染色观察血肿周围炎性细胞浸润 ,免疫组化方法观察NF -κB表达。结果 脑出血组血肿周围炎细胞浸润在 6h出现 ,4 8h达高峰 ,并持续到 7d。NF -κB阳性细胞于术后 1h开始增多 ,4 8h达高峰 ,并持续到 7d。假手术组未见血肿及明显炎细胞浸润 ,可见少量NF -κB阳性细胞。结论 脑出血大鼠脑内NF -κB的表达与出血后水肿规律存在时间上的相关性
【Abstract】 Objective To observe the expression and significance of NF-κB in rat brain after experimental intracerebral hemorrhage (ICH).Methods The male adult Sprague-Dawley rats were randomly divided into model group(42 rats) and sham group(6 rats).According to different time,the model group was divided into 7 sub groups (1 h,3 h,6 h,24 h,48 h,72 h,7 d),each one included 6 rats.Brain edema law was observed by dry-wet method,inflammatory cells infiltrating round hematoma by HE staining,and NF-κB expression by immunohistochemistry.Results After ICH,inflammatory cells infiltrating appeared at 6 h in perihematoma,reached maximum at 48 h,and continued to 7 d.NF-κB positive cells began to increase at 1 h,reached maximum at 48 h,and continued to 7 d.In the sham group,there was no hematoma and inflammatory cells infiltrating round hematoma,but there was little NF-κB positive cells.Conclusion There is the correlation in time between NF-κB expression in rat brain and brain edema after ICH.
