电脉冲激活对体外成熟猪卵母细胞卵裂率和囊胚率的影响
Effects of Electrical Activation on the Rates of Cleavage and Blastocyst of Porcine In Vitro Maturation Oocytes
【摘要】 探索了电脉冲结合细胞松驰素B(CB)、放线菌酮(CHX)以及6-二甲基嘌呤(DMAP)等化学物质,对猪卵母细胞激活后发育的影响。在电脉冲激活时,0.6 kV/cm和1.3 kV/cm,80 μs, 1次电激活后,(1)用2 mmol/L DMAP处理6 h的卵裂率分别为(60.39±6.71)%和(60.35 ±8.95)%,差异不显著(P >0.05),囊胚率分别为(9.51±5.93)%和(14.53 ±3.38)%,差异显著(P <0.05);(2)用7.5 μg/mL CB +10 μg/mL CHX处理6 h的孤雌胚,卵裂率[(41.82±10.76)% vs(58.50±8.33%)]和囊胚率[(8.91±4.37)% vs(14.53±3.38)%]均存在显著差异(P <0.05)。卵母细胞经1.3 kV/cm、80 μs和单次电脉冲激活后,分别用7.5 μg/mL CB、10 μg/mL CHX、2 mmol/L DMAP、7.5 μg/mL CB +10 μg/mL CHX和7.5 μg/mL CB +2 mmol/L DMAP处理2~8 h,结果表明:CB 处理4 h,CHX、CB + CHX和CB + DMAP处理6 h,DMAP处理8 h的卵裂率和囊胚率高于其它时间处理组。在电脉冲/CB、CHX、DMAP、CB + CHX和CB + DMAP 5种激活方法中,CB+CHX和CB+PMAP的卵裂率和囊胚率分别为(78.46±8.46)%、(33.13±7.84)%和(75.84±10.61)%、(39.12±7.15)%,囊胚率显著高于其它各组(P <0.05)。
【Abstract】 The effects of electrical pulses combined with CB(cytochalasin B), CHX(cycloheximide), or DMAP(6-dimethylaminopurine) on the activation of porcine oocytes were investigated. Under two different electrical pulses, 0.6 and 1.3 kV/cm, the percentages of activated oocytes developed to 2-cell stage ((60.39±6.71)% vs (60.35±8.95)%) were similar (P > 0.05) after electrical stimulation with direct current for 80 μs followed by 2 mmol/L DMAP treatment for 6 h, while the percentages of embryos developed to the blastocyst stage ((9.51±5.93 )% vs (14.53 ±3.38) %) were significantly different (P < 0.05). If the electrical stimulation was followed by CB 7.5 μg/mL) pulse CHX 10 μg/mL treatment for 6 h, the percentages of parthenogenetic embryos developed to 2-cell ((41.82±10.76 )% vs (58.50±8.33) %) and to blastocyst stage ((8.91±4.37) % vs (14.53± 3.38) %) were significantly increased along with the increase of the electrical pulse. When fixing the electrical pulse at 1.3 kV/cm for 80 μs, followed by 5 different treatments (CB, CHX, DMAP, CB+CHX, and CB+DMAP), the percentages of cleavage and blastocyst were (78.46±8.46) % and (33.13±7.84 )% with CB + CHX treatment and (75.84±10.61)% and (39.12±7.15)% with CB + DMAP treatment, respectively, which were significantly higher (P < 0.05) than that with other treatments.
