脂肽生物表面活性剂产生菌的筛选

【摘要】 从220个采自大庆油田和胜利油田的油土样和油水样中,经富集培养,血平板和油平板分离,液滴坍塌法、排油法和表面张力法进一步复筛,筛选出了8株产生物表面活性剂的优良菌株。经过薄层层析(TLC)和红外光谱分析(FT-IR)鉴定,有4株菌产脂肽表面活性剂,其中59号菌株最好,其表面活性剂可以把表面张力降到31mN/m而且具有良好的乳化活性。

【Abstract】 Microorganisms capable of producing biosurfactants could be isolated by a series of step including enrichment culture, hemolytic activity assay on blood agar plates, hydrolyzing oil activity assay on oil agar plates and oil displacement activity etc. Then the fermentation supernatant of first-isolated strains was secondly screened by a drop-collapsing method and surface tension method. Eight strain bacteria were isolated with higher surface activity from 220 samples of oil-contaminated soil and water collected from Daqing Oil Field and Shengli Oil Field. The results of TLC and IR analysis showed that biosurfactant of four strain bacteria was lipo-peptide and strain 59 was the best one among them.
