维生素E和槲皮素对多氯联苯引起的鸡胚肝细胞损伤的缓解作用

【摘要】 利用体外培养的鸡胚肝细胞研究抗氧化剂维生素E (vitamin E)和槲皮素(quercetin)对多氯联苯(polychlorinated biphenyls, PCB)引起的肝细胞损伤的缓解作用。取14 d的艾维茵鸡胚的肝脏,用胶原酶消化分离成单个肝细胞并进行培养。实验分对照组、PCB组(剂量为0.01~10 μg/mL)、维生素E或槲皮素组(剂量为0.1~10 μg/mL)和维生素E或槲皮素+PCB(10 μg/mL)组。药物处理48 h后用四甲偶氮唑蓝(MTT)法测定肝细胞的存活率。结果显示:在无血清培养液中鸡胚肝细胞呈多角形,成片生长,可存活1周以上。不同剂量的PCB组均可抑制肝细胞的增殖并引起细胞的部分死亡,高于0.1μg/mL的PCB能引起肝细胞明显死亡;维生素E和槲皮素组的肝细胞保持与对照组相似的形态,与PCB共同处理组中10μg/mL的维生素E或槲皮素可显著抑制PCB引起的细胞损伤,使细胞保持比较完整的外形,但剂量较低时作用不显著。结果表明离体培养的鸡胚肝细胞可作为肝损伤的细胞模型,PCB对培养的肝细胞的损伤及抗氧化剂维生素E和槲皮素对它的缓解作用提示, PCB对肝细胞的损伤可能是通过诱导细胞膜发生脂质过氧化而导致细胞的死亡。

【Abstract】 In vitro chicken embryo hepatocytes were adopted to evaluate the antioxidizing effects of vitamin E and quercetin on polychlorinated biphenyls (PCB)-induced toxicity. The hepatocytes were dispersed by collagenase digestion from 14 d Avian chicken embryos and cultured in serum-free medium 199. The cultured cells were challenged with PCBs, vitamin E or quercetin (0.1~10 μg/mL) alone or in combination for a period of 48 h. The morphology and death rate of the in vitro hepatocytes were monitored. At the end of culture the hepatocyte viability was determined by MTT method. The results showed that PCBs caused a dose-dependent hepatocyte death. And more than 10 μg/mL dose PCBs could significantly inhibited hepatocyte proliferation and cause hepatocyte death. The itamin E or quercetin alone maintained hepatocyte morphology that was similar to the control group. As antioxidants, the vitamin E or quercetin (10μg/mL) significantly attenuated the toxic effects of PCBs on the cultured hepatocytes. The above results indicated that vitamin E or quercetin might antagonize the toxic effect of PCBs via antioxidizing effects on in vitro cultured chicken embryo hepatocytes.
