两种方法对螺旋藻DNA和RNA含量的测定
DETERMINING CONTENT OF DNA AND RNA IN Spirulina WITH TWO METHODS
【摘要】 以鄂尔多斯高原碱湖的钝顶螺旋藻S1、鄂尔多斯螺旋藻S2、引进的钝顶螺旋藻S3和极大螺旋藻S4为材料,采用两种方法分别测定了其DNA和RNA的含量。结果如下:改良二苯胺法测定的DNA含量为1.03μg/mgDW~1.68μg/mgDW;紫外分光光度法测定的为5.86μg/mgDW~10.03μg/mgDW。两种方法测定的DNA含量排列均为S2>S4>S1>S3。苔黑酚法测定的RNA含量为14.36μg/mgDW~22.56μg/mgDW;紫外分光光度计法测定的为17.67μg/mgDW~39.53μg/mgDW。两种方法测定的RNA含量排列均为S2>S1>S3>S4。无论DNA还是RNA,紫外分光光度法测定的值高。
【Abstract】 The content of DNA and RNA in Spirulina(Arthrospira)platensis(S1),S.(A.)erdosensis(S2)from alkaline lake in Erdos Plateau and the introduced S.(A.)platensis(S3)and S.(A.)maxima(S4)were determined with two methods respectively. The results showed that the content of DNA in Spirulina determined by means of ameliorative diphenylamine was 1.03~1.68μg/mgDW;That determined by ultraviolet absorption was 5.86~10.03μg/mgDW. Both methods showed following tendency:S2>S4>S1>S3.The content of RNA in Spirulina determined by means of 3,5-Dihydroxytoluene was 14.36~22.56μg/mgDW.That determined by ultraviolet absorption was 17.67~39.53μg/mgDW. Both methods showed following tendency:S2>S1>S3>S4.The content whether DNA or RNA determined by means of ultraviolet absorption is higher.
