马鞭草C部位使人绒癌JAR细胞阻滞于G2/M期并诱导细胞凋亡

【摘要】 目的:观察马鞭草C部位(VerbenaofficinalisC)对人绒癌JAR细胞周期分布和凋亡的影响,探讨其诱导凋亡机制。方法:采用MTT比色法测定马鞭草C部位对JAR细胞增殖的影响;流式细胞仪检测其对JAR细胞周期分布和细胞凋亡的影响;RT鄄PCR检测药物作用前后Bax和Bcl鄄2mRNA表达变化。结果:马鞭草C部位抑制绒癌JAR细胞增殖,并呈明显时间和剂量效应关系。流式细胞仪结果显示,与对照组比较,40g/L马鞭草C部位处理48h后,使JAR细胞G2/M期增加154.3%,S期比例降低41.6%;JAR细胞凋亡率增加6.74倍。不同剂量马鞭草C部位处理JAR细胞48h后,Bax表达水平增加,Bcl鄄2表达水平下降,并呈剂量依赖性。结论:马鞭草C部位阻滞绒癌JAR细胞于G2/M期,通过改变Bax和Bcl鄄2表达,诱导JAR细胞凋亡。

【Abstract】 Objective: To investigate the effects of Verbena officinalis C on cell cycle arrest and apoptosis in Human ChoriocarcinomaCell JAR cell line. Methods: Firstly, JAR cells treated with Verbena officinalis C were determined the proliferation-inhibition ratio by MTT method. Secondly, flow cytometry(FCM) was employed to show phase change of cell cycle and apoptosis. Thirdly, the expressions of Bax and Bcl-2 were determined using RT-PCR. Results: Verbena officinalis C inhibited proliferation of JAR cells in a dose and time-dependent manner. After JAR cells treated with 40 g/L Verbena officinalis C for 48 h, cells in the G2/M phase increased 154.3% but cells in the S phase decreased 41.6% in comparison with control. Apoptosis ratio was 6.74 times more than control. After JAR cells treated with Verbena officinalis C, the expression of pro-apoptotic gene Bax increased but anti-apoptotic gene Bcl-2 decreased in a dose-dependent manner. Conclusion: Verbena officinalis C inhibits JAR cells proliferation, blocks JAR cells in G2/M phase and induces apoptosis via changes of Bax and Bcl-2 expression level.
